Pseudomonas aeruginosais a Gram-negative opportunistic human pathogen responsible for a number of healthcare-associated infections. Assessing single-cell contributions ofP. aeruginosato antibiotic resistance or virulence is a current challenge, because mechanistic behavior is conventionally inferred from ensemble cultures, thus averaging effects over a population. Here, we develop and characterize a device that can capture arrays of singleP. aeruginosacells in individual micropores to study their spectroelectrochemical behavior. An array of individual micropores is fabricated with dimensions designed to capture singleP. aeruginosacells in a vertical orientation. Upon application of an electrochemical potential waveform to the bottom Au layer, three characteristic fluorescence behaviors are observed from trapped cells that are sensitive to external conditions. The device developed here enables the study of parallel collections of single bacterial cells and will facilitate efforts to elucidate methods of bacterial communication and multidrug resistance at the single-cell level.
