Irradiation of peptides dissolved In glycerol, ethylene glycol, 1, 4-butanedlol, or 1, 5-pentanedlol by a high-energy (klloelectronvolts) beamof Xe atoms results In the formation of several adducts having masses greater than the MH+ Ion, which were characterized by mass spectrometry In the pos-itive Ion mode. The most abundant of those adducts corre-sponded to the addition of 12 u to the protonated molecular Ion (MH+) of the peptides. Several other adducts were also visible In the mass spectrum. The corresponding thiols (a-thloglycerol and dithlothreltol/dlthloerythrltol) demonstrated poor affinity for the adduct formation with the peptides. The adduct formation was facilitated In dilute and In basic solutions of the peptides and was reduced drastically In acidic solutions. Prolonged irradiation also favored the adduct formation. The mass spectrometry/mass spectrometry spectra obtained by using the B/E Hnked-fleld scan mode revealed that the slte (s) of attachment of glycerol to the peptide was the most basic site In the molecule—the free N-termlnus amino group or the primary amine group of a basic (Lye, Arg) amino acid residue. Electrophilic attack of the Ionized glycerol and Its oligomers onto the N atom of the highly basic amino group and subsequent fragmentation of the resulting adducts appear to be the favored mechanism for the formation of the adducts. Evi-dence of the addition of up to at least three molecules of glycerol was revealed.
