pGlu-His-Pro-NH2 (TRF) is shown to be a potent stimulator of PRL secretion in the estrogen-progesterone-pretreated male (EP) rat. A series of structural analogues of pGlu-His-Pro-NH2 were found to have comparable potencies to stimulate both PRL and TSH in the EP rat. Using pGlu-His-Pro-NH2 as a standard, the amount of PRL releasing activity in various hypothalamic extracts, as determined in the EP rat, is in agreement with the amount of TSH releasing activity as measured by a mouse TRF assay, but is not in agreement with the amount of TSH releasing activity determined by measurement of plasma TSH levels in the EP rat as the assay animal. This discrepancy is best explained by a differential sensitivity of the two methods, to TSH release inhibiting substances such as somatostatin. The PRL releasing activity as well as the TSH releasing activity of hypothalamic extracts is destroyed by incubation in rat serum.
