Leukemias originate from stem cells or in the myeloid and lymphoid lineages, the acute or chronic forms having quite distinct clinical patterns. For example, chronic leukemia usually has an insidious onset, while acute leukemias usually manifest an abrupt onset. In addition, the acute leukemias (except myeloid) have aa peak of incidence in children around the age of two to three years; the chronic leukemias tend to have a greater incidence in older patients. These clinical features suggest different mechanisms in tumor development between acute and chronic leukemias mediated by different, somatically acquired genetic changes.A characteristic of leukemias is the presence of specific chromosome abnormalities considered to be involved in tumor development (proven experimentally in some cases and implicated by association in others). Molecular studies of genes adjacent to the breakpoints of these chromosome aberrations and studies on the function of their gene products have elucidated the general role of chromosome translocations in tumor initiation and progression.
