Extracellular purine nucleotides such as ATP and its derivatives play important roles in the modulation of T lymphocyte functions. P2X receptors are ATP-gated cation channels that permit the influx of calcium and sodium into and the efflux of potassium out of the cell. In recent years evidence has accumulated that P2X receptors positively reinforce the activation of T lymphocytes upon T cell receptor (TCR) ligation by increasing cytosolic calcium levels within the cell. Although this concept has gained wide acceptance, the precise mechanisms by which P2X receptors contribute to TCR signaling remain unclear. This review summarizes current knowledge about the effects of P2X receptors on the development and functions of T lymphocyte subsets, and focuses especially on their role in the context of TCR signaling.
