Emilio Rivas,* Frangoise Reiss-Husson, and Marc le Maire5 abstract: Reaction centers purified from Rhodopseudo-monas spheroides (wild-type and R 26 strains) were charac-terized in detergent solutions by sedimentation velocity and sedimentation equilibrium analysis in detergent buffers con-taining various amounts of H2, sO. In therange of detergent concentrations studied (0.5-2.0 mg of dodecyldimethylamine TV-oxide per mL), the detergent-reaction center complexes are monodisperse; sedimentation equilibrium experiments in H20-H2180 buffers revealed particle weights of 138 900 and 140700 for the R 26 and wild-type reaction centercomplexes, respectively. Under these conditions 0.67 and 0.58 g of do-decyldimethylamine TV-oxide were bound per g of protein in R 26 and wild-type preparations, respectively, as determined by chromatography using 14C-labeled detergent.
