The hydrolytic rate of p-nitrophenylphosphorylcholine by phospholipase C [EC 3.1.4.3] is highly accelerated in a medium containing sorbitol. Glycerol is also effective in increasing the hydrolytic rate but does not increase it as much as sorbitol. A convenient spectrophotometric assay method for phospholipase C using p-nitrophenylphosphorylcholine together with sorbitol (60%, wt/wt) was proposed. This method was very useful for studying the metal and detergent requirement of phospholipase C. The results obtained using this method suggest that phospholipase C of Clostridium perfringens is a metalloenzyme possibly requiring Zn2+ and that no type of detergent has any activating effect on the enzyme itself.
