Failure of oocytes to fertilize in vitro may be due to several causes, including defective gamete interaction or inability of oocyte cytoplasm to form male and female pronuclei. The situation is particularly confusing in those cases in which results of the semen analyses are within normal limits, and the oocytes appear mature and healthy. To distinguish between defective gamete interaction and oocyte cytoplasmic dysfunction, it would be useful to remove the zona pellucida barrier to sperm penetration of the oolemma. The subsequent appearance of pronuclei would indicate adequate oocyte cytoplasmic functions and imply that fertilization failure was a defect of gamete interaction. Mild trypsinization treatment to weaken the zona barrier is a possible approach to understanding the problem since such treatment is routinely applied to cells in culture without evidence of significant membrane damage. We report here the use of mild trypsinization to improve fertilization of oocytes from three patients with a history of fertilization failure.
