Rapid sampling and analysis of the neurotransmitter dopamine present in the extracellular fluid of rat brain have been achieved by use of microdialysis. Concentrated 0.5-.mu.L samples are obtained by using a perfusion flow rate of 210 nL/min. A 5-min analysis is performed with a smallbore (1.0 mm i.d.) liquid chromatographic column and electrochemical detection. The retention time for dopamine is 3 min. The time scale of the analysis is appropriate for the study of acute pharmacological and behavioral manipulations. The chromatographic system is evaluated on the basis of detection limit and dynamic response to concentration changes in the sampled medium. The performance of the system is characterized in vitro and in vivo. The system can routinely measure 20 nM dopamine in 0.5-.mu. L samples.
