Deoxyhypusine synthase catalyses the formation of deoxyhypusine [Nε-(4-aminobutyl) lysine]. This modified lysine residue is the key intermediate in the posttranslational synthesis of hypusine [Nε-(4-amino-2-hydroxybutyl) lysine] in eukaryotic initiation factor 5A (eIF5A) 1 (reviewed in Park et al., 1993, 1997). Hypusine and eIF5A occur in all eukaryotes examined, and in certain archaea, but not in eubacteria (Gordon et al., 1987; Schümann and Klink, 1989; Bartig et al., 1990; Park et al., 1993, 1997). The essential nature of hypusine for eukaryotic cell proliferation has been strongly implicated (Byers et al., 1992; Park et al., 1993, 1997; Byers et al., 1994) and recently established in experiments in which inactivation of the genes for eIF5A (Schnier et al., 1991; Schwelberger et al.,
