METHODSMale Sprague-Dawley rats1 weighing between 185 and 250 g were fed a basic diet containing 0.287 mEq sodium per gram and 0.207 mEq potassium per gram (GL Baking Co., Frederick, Md.) and tap water. Where indicated, either a 2% potassium chloride solution or a 1% sodium chloride solution was substituted for tap water. After 2 days of equilibration in individual metabolic cages designed to separate urine from feces, a 24-hr balance was determined in control rats as follows: fluid and food intake were measured, and the amounts of sodium and potassium consumed were calculated; urine volume and urine osmolality were measured, and sodium and potassium excretion were determined. Similar 24-hr balance studies were obtained in potassium-loaded rats 1, 2, or 3 weeks after substitution of 2% potassium chloride for tap water. Control and potassium-loaded animals were anesthetized with sodium pentobarbital(50 mg/ml), 50 mg/kg intraperitoneally, for the determination of inulin clearance and plasma volume immediately after the 24-hr balance studies. Both femoral veins were catheterized and a flanged catheter (PE-90) was placed in the bladder through a ventral incision and secured with a purse-string suture. A modification of a previously described method was used for the determination of plasma volumes (37). Briefly, a precise volume of 1251-labeled human serum albumin(Albumutope, ER Squibb & Sons, New Brunswick, NJ) diluted in 0.85% saline was given intravenously using a micrometer syringe, and the catheter was flushed with 0.2 ml of saline. The total radioactivity of the administered dose was determined by delivering at least 4 identical volumes into counting tubes. The range of individual samples was less than 3%.
