Ethylene production wasstimulated severalfold during thehypersensitive reaction ofSamsunNN tobacco totobacco mosaic virus (TMV). Exogenous methionine orS-adenosyhnethlonlne (SAM)didnotincrease ethylene evolution fromhealthy orTMV-infected leaf discs, although both precursors weredirectiy available forethylene production. Thisindiates thatethylene production isnotcontrolled atthelevel ofmethionine concentration oravailability, noratthelevel ofSAM production or concentration. Incontrast, 1-aminocyclopropane-1-carboxylic acid (ACC) stimulated ethylene production considerably. Thus, ethylene production is primarily limited atthelevel ofACCproduction. Theregulation ofethylene production during thehypersensitive reaction toTMV wasfurther studied bydetermining thetimecourseofthe concentrations ofmethionine, SAM,andACC,aswell asthecourse of their invivo conversion rates. Endogenous concentrations ofmethionine andSAMremaned unaffected until late ininfection. Onthecontrary, the peakinethylene production nearthetime oflocal lesion development was preceded byalarge increase inACCproduction. Asa result ofthis increase, ACCaccumulated intheleaf tissue. Only after local lesions becamevisible, thecapacity toconvert ACC intoethylene increased severalfold, associated with asharp decrease inACCcontent andalarge increase inethylene production. Ethylene production intobacco leaves reacting hypersensitively toTMV isthusregulated atthelevel ofboththeproduction ofACCandits conversion toethylene.
