Crude methanol extracts of posterior salivary glands of Eledone, a Mediterranean eight-armed cephalopod, were subjected successively to chromatography on alkaline alumina (or to absorption on serum albumin), to ion-exchange chromatography, and to countercurrent distribution. The active constituent of these extracts, the polypeptide eledoisin, was thus obtained in a pure form. Total and partial acid hydrolysis, fission with trypsin and chymotrypsin, and end-group determination experiments demonstrated that eledoisin is an endecapeptide having the following amino acid composition and sequence Pyr-Pro-Ser-Lys-Asp(OH)-Ala-Phe-lle-Gly-Leu-Met-NH2.
