Our understanding of lymphatic biology has increased tremendously in recent years. The primary role of lymphatic vessels is to regulate fluid dynamics within a given tissue. Lymphatics recover interstitial fluids released from blood vessels, transport it to lymph nodes and recirculate the fluid back to the venous system. The lymphatic vascular system develops following that of the circulatory system with initial lymphatic vessels sprouting from veins. Great strides have been made in investigating the cellular and molecular bases of lymphangiogenesis, the specification of lymphatic endothelial cells, and their maturation and differentiation. Failure of the lymphatic system to adequately control fluid results in lymphedema and swelling. Abnormal lymphatic vessel formation results in pathologies such as lymphangiomatosis or lymphatic malformations. Lymphatic vessels also serve many other functions that are important for normal homeostasis including lipid transport and cell trafficking. Finally, metastatic tumor cells also use this preformed lymphatic “highway” to gain access to distant organs.This special issue of Microvascular Research focuses on several emerging areas of high interest within the field of lymphatic biology including mechanisms involved in the development of the lymphatic system from the blood vascular system as well as lymphatic specification, maturation, and valve development. This issue provides ten review articles that focus on both the development of the lymphatic system and the pathologies that result when these vessels are dysfunctional or usurped by cancer cells.
