Gangliosides that bind cholera toxin can be detected by the direct binding of 125I-labeled toxin to thin-layer chromatograms followed by autoradiography. The method is very sensitive: 0.1 ng of GMI (70 f[femto]mol) can be detected on chromatograms of total lipid extracts of cells. The method may be useful in defining the receptors of other toxins or proteins such as some lectins, antibodies and hormones that bind carbohydrates.
