Hepatic cell nuclei were studied in radioautographic preparations of livers from 230 g male rats killed 2 and 28 hr. after injection of tritiated thymidine. Two hours after injection, labelled nuclei were of polyploid size, with a majority in the octoploid size range. Twenty-eight hours after injection, labelled nuclei were predominantly in the tetraploid-size range, with a few in the diploid and octoploid ranges. The distribution was the same as that of nuclei in the total cell population. The labelling index was 1.0 [plus or minus] 0.1%, 2 hr. after injection, and increased significantly, to 1.9 [plus or minus] 0.39%, 28 hr. after injection, while the mitotic indices were 0.05 [plus or minus] 0.03% and 0.03 [plus or minus] 0.001% of cells, respectively. Tetraploid-sized nuclei formed a large proportion of the reproductive cell population. They incorporated tritiated-thymidine and enlarged to octoploid size, subsequently dividing to produce daughter tetraploid nuclei. New binucleated cells were formed through a process involving new DNA synthesis, and binucleated cells were engaging in DNA synthesis. A few nuclei may be shifting from the diploid to the tetraploid state through the formation of binucleated cells.
