METHODSExperiments were conducted in 24 male guinea pigs (body mass ca. 450 g). The animals were prepared for the experiments as described in detail previously. 4 Briefly, they were implanted with an intraperitoneal thermode and intrahypotha-lamic thermocouple, allowed at least one week for recovery from surgery, and habituated to the experimental conditions. During the experiments, the guinea pigs were lightly restrained. IPH was accomplished by perfusing water through the thermode (perfusion rate: 50 ml/min; inflow water temperature: 45 C; perfusion duration: 80 min). NTX-hydrochloride (Research Biochemicals International, Natick, MA; 50 or 0 μmol/kg) in pyrogen-free saline (PFS; 1 ml/kg) was injected subcutaneously immediately before the beginning of IPH. In control experiments, NTX was similarly injected at 50 or 0μmol/kg, but no IPH was performed. Hypothalamic (T h) and ear skin (T sk) temperatures were monitored, and the heat loss index (HLI) was calculated according to Székely's 5 formula: HLI=(T sk-T a)/(T h-T a), where T a is the ambient temperature (in our case, ca. 24 C). The value of HLI varies from 0 (full vasoconstriction) to 1 (full vasodilation). The HLI has been successfully used before for evaluation of the effector thermoregulatory responses in the guinea pig's ear. 6, 7RESULTSThe initial thermal state of the guinea pigs was characterized by normothermia (the average basal T h for all experiments was 38.3±0.2 C) and moderate skin vasoconstriction (the mean HLI was 0.49±0.02).
