It has recently been reported that when perilymph is removed from scala tympani of guinea pig cochlea, the tuning curves of auditory nerve ganglion cells are seriously affected. This suggests that measurements of basilar membrane vibration under similar conditions may also be affected and not representative of the normal cochlea. Hence the discrepancy known to exist between neural tuning curves and basilar membrane tuning curves could be explained. We have confirmed the existence of this effect on neurons, but we present two kinds of evidence that it is not due to a basilar membrane change. First, the amount of perilymph which must be removed is greater than that routinely removed for mechanical measurements. Second, filling the scala with isotonic sucrose produces the same change.
