The synthesis of the major histone proteins in mammalian cells is tightly coupled with DNA synthesis. The concentrations of the histone mRNAs vary 30-50-fold during the cell cycle, 2 and the changes in mRNA concentration occur very rapidly in parallel with changes in DNA synthesis. The decrease in histone mRNA concentrations which normally occurs following DNA synthesis can be mimicked by treating cells with inhibitors of DNA synthesis 3, 4. Recent studies in our laboratory and others studying control of histone mRNA metabolism in mammalian cells have revealed that there are multiple control steps in the metabolism of histone mRNA. The major control steps are post-transcriptional, and recent studies have led to a start towards understanding the molecular basis of this novel control mechardsm.
