The model postulates that random X inactivation in eutherians has evolved from an ancestral paternal X inactivation which has been retained in marsupials. It proposes that during ♂ meiosis in both groups the X-chromosome undergoes a directed genetic change. A controlling element is introduced into the X, probably by the Y. Then, at a certain point in the very early development of eutherian ♀♀, this controlling element is excised and later re-inserted at random into one of the 2 X-chromosomes, thus setting up random X inactivation. Once re-inserted, it remains fixed in that chromosome. The adequacy with which this model accounts for data at present available is examined, and possible experimental tests are suggested.
