Nicotine is a major constituent of tobacco and tobacco smoke; it is extensively metabolized by man and laboratory animals to give a large number of metabolites. Nicotine-.DELTA.1''(5'')-iminium ion, either as the diperchlorate or the hydrochloride, is rapidly converted into continine by a hepatic cytoplasmic enzyme. Incubations in an atmosphere of N indicated that the O incorporated into continine is derived from H2O. Following the i.p administration of nicotine-.DELTA.1''(5'')-iminium ion to guinea pigs or rats, cotinine, and nicotine-1''-N-oxide were found in the wine. Nicotine-.delta.1''(5'')-iminium ion is evidently an intermediate in the metabolic conversion of nicotine to cotinine.
