The X-ray survival of haploid yeast is shown to consist of 2 components: an initial exponential (LD50 = 3100 r) which represents the survival of interdivisional cells, and a radioresistant (LD50 = 60,000 r) multiple hit (target number ca. 1000) component which represents the survival of cells in the process of division. The ability of clumps of cells, occasional diploid cells, and a distribution of radiosensitivities to account for this radioresistant component are eliminated. Evidence that budding cells represent the radioresistant component is twofold: 1. Survival curves in which survival of single and budding cells from a growing culture is followed microscopically show budding cells to be more radioresistant. 2. Suspensions of resting cells (starved 2-5 d., 30[degree]C, without usable nitrogen) have few buds and few radioresistant members. When such cells are fed, the percentage of fresh buds is correlated with the rising level of the radioresistant component.
