Staphylococcus aureus (S. aureus) possess a surface component, protein A, which has the unique property of reacting with the Fc portion of the IgG molecule of most mammalian sera. 1"" Specifically with regard to this property, we studied the localization of protein A on the cell surface of the Cowan I and Wood 46 strains, and HL-87, 4'an isolate of the former which lacks protein A. S. aureus, Cowan I and Wood 46 strains, which were kindly supplied by Dr. Y. Arai, Saitama Medical School, were maintained in our laboratory. Five protein A-deficient strains were isolated from the Cowan I strain by a modification of the method of Masuda et a/. 61 One of the isolates, HL-87, was examined together with the Cowan I and Wood 46 strains. The bacterial cells of about 10" colonyforming units/ml were treated with lysostaphin (5 units/ml, Sigma Chemical Co., USA) and centrifuged at 2,300 g to remove the cell debris. The supernatant was assayed by the hemagglutination test described by Spika et al. e) The titer was expressed as the reciprocal of the highest dilution showing visible hemagglutination.
