Oestradiol administered at the same time as PMS appeared to inhibit PMS-induced ovulation in 24-day-old rats killed 72 h after treatment. However, autopsy at 48 h showed that a premature ovulation had occurred. Premature ovulation did not occur when the oestrogen was injected 24-52 h after PMS. A dose of 2.5-80 mu g oestradiol induced premature ovulation, but one of <2.5 mu g had no effect. Oestradiol interacted with progesterone in the induction of premature ovulation. However, the action of progesterone on LH release was dependent on a preceding exposure to an adequate level of oestrogen. Oestradiol had no effect on ovarian sensitivity to an ovulatory dose of 80 mu g of LH. The premature ovulatory response induced by the injection of oestradiol at the same time as PMS was blocked by pentobarbital from 27-32 h, but not at 24 or 33 h. These results suggest that the injection of oestradiol at the same time as PMS advances PMS-induced ovulation by means of an effect upon the CNS mechanism controlling the release of LH.
