During the past few years various approaches have been made to elucidate the genetic coding problem. The most successful approach began with the observation by Nirenberg and Matthaei (1961) that poly U stimulates the synthesis of polyphenylalanine in a bacterial subcellular system. Another method consists of comparing amino acid exchanges in specific proteins with nucleotide alterations produced in the genetic material by specific mutagens. This has been accomplished with mutants of tobacco mosaic virus (TMV)(Wittmann, 1959, 1960, 1962, 1964; Tsugita and Fraenkel-Conrat, 1960, 1962; Funatsu and Fraenkel-Conrat, 1964) and of tryptophan synthetase (Yanofsky, 1963).
