Abstract The present study examines the reaction leading to the formation of thrombin from its precursor, prothrombin. The data demonstrate that the amount of thrombin generated from a mixture of prothrombin and the "prothrombin-converting principle," derived from the interaction of activated Stuart factor and proaccelerin in the presence of phospholipid and calcium ions, is directly related to the amount of prothrombin initially present. The rate of generation of thrombin, however, is a function of the concentration of the "prothrombin-converting principle," and is greatly retarded at a temperature of 0 ° C. These data provide experimental evidence that the activation of prothrombin is enzymatic, a view long held by many investigators. The cations calcium or strontium must be present for thrombin to evolve from prothrombin under the conditions used in this study.
