Materials and MethodsThe origins of the B-16 melanotic melanoma cell line used in these experiments have been previously described (Kreider et al., 1973). The amelanotic clone was isolated from the stock B-16 tumor maintained by the Jackson Laboratory, Bar Harbor, Maine. A high molecular weight chondroitin of varying sulfate content, predominantly 4-sulfate, is a major product of the cloned lines; this poly-saccharide was not detected in the iris cultures. Heparitin sulfate was present in all cells but appearedto be somewhat higher in content for the tumorigenic clones. chip method (Martin, 1973); the resulting line produces only very small amounts of melanin. All cells were routinely propagated in 16-oz prescription bottles and fed with mini-mum essential mediumwith Earle's salts, 10% heat-inactivated fetal calf serum supplemented with nonessential amino acids, sodium pyruvate, and twice the usual concentration of vita-mins.
