Relative concentrations of inftctive wound-tumor virus (WTV) in extracts from sweet clover tumors (Melilotus officinalis(L.) Lam.) and insect vectors (Agallia constricta(Van Duzee)) were measured by using an experimentally derived regression curve relating the concentration of virus injected into vectors to the wound-tumor soluble antigen (WTSA) titer reached in them 21 days later. The WTSA titers were determined by a precipitin ring-time test and were relative to the titer of a standard WTSA preparation.After injection of two different concentrations of WTV into groups ofA. constrictathe WTSA attained a plateau level earlier in the insects injected with the higher concentrations of WTV. Later the same WTSA plateau was attained in insects injected with the lower concentrations.After a 1-day acquisition access period, the WTSA and WTV concentrations inA. constrictarose steeply from day 6 to day 27 and then decreased slightly. The virus concentration increased 1000-fold between day 7 and day 30 after acquisition, and between day 30 and day 45 it decreased to about one-tenth of the peak concentration. The drop in the WTV and WTSA concentration was not caused by a hypothetical higher mortality among viruliferous than nonviruliferous insects.
