Crude herpesvirus suspensions were fractionated by the cesium chloride density gradient sedimentation method into enveloped and naked virus particles. Particles without envelopes were found to be almost non-infectious, whereas fractions containing enveloped particles were highly infectious. It appears, therefore, that the envelope plays an essential role in herpesvirus infection.
