White adipocytes have significant storage capacity. Accordingly, they are subject to volume changes of greater amplitude than those usually observed in other cell types after osmotic shocks. Obesity worsens fat cell hypertrophy, and it is known that metabolic functions (storage and mobilization of lipids and hormonal secretion) are subject to change during the development of adipose tissue. 1 Since obesity is characterized by nutritional and hormonal abnormalities, it is not known whether the changes observed in enlarged fat cells of obese subjects are solely attributable to these abnormalities. However, in nonobese healthy subjects, adipose cell volume also increases currently by 20 to 30 times (a 3-fold increase in diameter is equivalent to a 33-fold increase, or 27-fold increase, in volume). When adipocytes of very different sizes are present at the same adipose location, little is known about fat cell metabolism as a function of cell size. Thus, the first aim of this work was to examine the same fat depot (in rats) so that we might determine whether several metabolic functions of adipocytes, independent of changes in nutritional or hormonal conditions, were subject to change with cell size. Two populations of adipocytes were isolated from epididymal adipose tissue (52 and 22× 103 µ3), and we examined enzyme activity and expression of several proteins involved in the main functions of adipose cells:(1) storage (glucose transporter Glut4 mRNA concentration, both activities and mRNA concentrations of lipoprotein lipase, LPL, and fatty acid synthase, FAS);(2) mobilization of lipids (in vitro lipolysis and expression of hormone-sensitive lipase, HSL);(3) hormonal production (leptin mRNA concentration).
