B iological methods, such as exp ression using bacteria, are useful. They are, however, not al2 ways app licable to the synthesis of pep tides with site2 specific modifications. Chem ical methods can be viable alternatives to those biological app roaches. Pep tides obtained by chem ical and biological means, as building blocks, offer a great potential to the synthesis of a wide variety of polypep tides. In 1991, Hojo and A imoto [1, 2] reported a method for polypep tide synthesis in which S 2alkyl pep tide thioesters (pep tide thioesters) were used as building blocks. Since then, imp rovements have been made to the thioester method [3—5]. The method is now useful in the synthesis of various types of polypep tides (Fig. 1) including phospho2 6] and glycop roteins 7].
