METHODSAdult male Wistar and Long-Evans rats under 6 months of age, usually weighing 300-450 g, were used. The animals had free access to standard laboratory chow and water until the time of the experiment. At that time, sodium pentobarbital was injected (50 “g/kg, ip) and surgical procedure begun.Surgical procedure. For experiments with colloidal tracer, both submaxillary glands and a length (about 1 cm) of the hilar end of each submaxillary duct were gently separated from the sublingual glands and ducts. One femoral vein was cannulated with polyethylene tubing filled with heparinized saline. For perfusion of the main excretory duct, the surgical procedure was similar to that described by Young et al.(20). The surgical sequence employed here involved cannulation, first, of the oral end of one submaxillary duct, using fine polyethylene tubing (Clay-Adams PE-10) beveled for easier insertion (18). No ligature was used to retain the oral cannula. Next, the ipsilateral submaxillary gland was exposed and separated from the adhering sublingual gland. The duct of the sublingual gland was separated from the submaxillary duct for a length of several millimeters and ligated. The trachea was cannulated. Finally, using loupes, the hilar end of the submaxillary duct was opened with iris scissors and a fine-tipped(80-p) polyethylene cannula was inserted distally into the lumen of the duct, for a distance of about 5 mm, and ligated in place. Vascular marking. Colloidal carbon was used as a
