MATERIALS AND METHODSThe cell lines, their growth, and cell cycle progression analysis by flow cytometric methods were described previously (Kung et al. 1990a). y radiation treatment protocols were also described previously (Sharma and Schimke 1989). Cytochalasin D (Sigma) was added 2-4 hours postirradiation to a final concentration of 2/xg/ml. Cell cycle analysis by flow cytometry was performed with the Cytologic Software package (Coulter Electronics), and cell frequency in each cell cycle phase represents the average value derived from five different DNA analysis models. Cell size was measured as forward angle light scatter (FALS), which correlates well with total cell protein content in fixed cells (Sherwood et al. 1988). All experiments employing cell synchronization used mitotic" shake-off" techniques (Kung et al. 1990a), inasmuch as various chemical and physical synchronization techniques may result in perturbation of the normal progression of the cell cycle.
