The relationship between transient peak plasma fluoride [F-] levels (s.c. injections) and lower but relatively constant levels (s.c. constant infusion) and the occurrence of disturbances in the enamel mineralization of the rat incisor as determined microradiographically was studied. The F- doses were administered for 1 wk and the animals were killed 2 wk later. Once-daily peak plasma F- levels of 10 .mu.M were uniformly associated with disturbances in the mineralization of the enamel, but once-daily peak levels of 5 .mu.M or twice-daily 5-.mu.M peaks were not. Relatively constant plasma F- levels, which averaged 3.3 .mu.M, were associated with an increased incidence of changes in enamel formation, and levels of 4.7 .mu.M consistently associated with disturbances in enamel mineralization. The rat was a better model for the study of human enamel fluorosis than previously believed, and slightly elevated but relatively constant plasma F- levels were more likely to be associated with the occurrence of fluorotic disturbances in the mineralization of enamel than were the rarer high and transient peak levels.
