(Refs 14, 15). Very little is known about the functions of the SIR proteins; for example, none of them appears to bind directly to the silent copy DNA 16. SIR2 appears to affect other aspects of chromosome structure, as a sir2 deletion mutant shows elevated levels of rDNA recombination 17. Interactions between SIR gene products have been inferred from studies showing suppression of sir mutations by multiple copies of a different SIR gene TM. The involvement of protein transacetylases and of histone H4 might suggest that silencing involves changes in histone acetylation; however, replacements of lysines in the amino terminus by arginines allows silencing, though alanine replacements turn on the silent gene 14, 15.
