After mutagenizing a recombinant between fowl plague virus (FPV) and virus N a temperature-sensitive (ts) mutant (ts 412) was isolated which carried a defect in the gene coding for the nonstructural (NS) proteins. This mutant was characterized in biological terms. The NS proteins are early proteins, but the lesion of its 412 is a late defect. At the nonpermissive temperature all viral products are synthesized in normal yields except for hemagglutinin and M protein which are produced in relatively low amounts. No particles are formed at 40.degree. C. Electron-dense inclusions were found under these conditions in the cytoplasm of chick embryo cells infected with ts 412. The structures may be accumulations of one of the NS proteins.
