The Effect of Environment on the Agar Yield and Gel Characteristics of Gracilaria sordida Nelson (Rhodophyta)

Gracilaria sordida Nelson (Rhodophyta), an agarophyte endemic to New Zealand, was grown under a range of photon flux density (PFD), temperature, and nitrogen levels under controlled conditions. Agar extracted from alkali-treated dried seaweed samples was then analysed for yield and quality using the standard industrial empirical parameters; gel strength, gel deformation and dynamic gelling temperature. Methods were developed to obtain this data from dried seaweed samples of about 1 g. Agar yield decreased with increasing water temperature between 10 °C and 30 °C, increased at nitrogen levels suboptimal for growth and was not affected by PFD. Gel strength increased with increasing temperature, and decreased with increasing PFD. Deformation was unaffected by temperature and PFD, but decreased with increasing nitrogen level. PFD and nitrogen altered the gelling temperature whereas growth temperature did not. Under certain conditions, cultured seaweed produced comparable yields and agar properties to those of harbour-grown plants and an agar gel strength within the range of commercial grade agars.
