Freshly disaggregated embryonic chick myogenic cells were cultured together with embryonic mouse muscle cells and with both untreated and actinomycin D treated mouse embryonic liver cells. Examination of muscle straps which developed in these cultures using autoradiography and histological staining to determine cell type confirmed that muscle cells from different organisms are incorporated into common multinucleated muscle units, but that non-muscle cells are excluded. Actinomycin D treated liver cells, however, are incorporated. The analysis of RNA and protein synthesis in these cells indicated a rapid and complete cessation of uridine incorporation and an almost complete cessation of amino acid incorporation. These results suggest the existence of a recognition process which is disturbed by the disruption of normal RNA and protein synthesis mechanism. A recognition factor at the cell surface is proposed.
