Two high-affinity Fe uptake systems are known in S. typhimurium, one utilizing Fe-enterochelin and the other utilizing ferrichrome. It was shown previously that expression of several elements of the Fe-enterochelin uptake system are regulated by the Fe content of the medium, with growth in high-Fe medium resulting in repression of enzymes of enterochelin synthesis and degradation and of the ability of whole cells to take up Fe-enterochelin. A mutant strain in which growth in high-Fe medium was associated with constitutive expression of: Fe-enterochelin uptake by whole cells; ferrichrome uptake by whole cells; synthesis of enterochelin; intracellular degradation of Fe-enterochelin; and synthesis of 3 major outer-membrane proteins (OM1, OM2 and OM3), was described. In the wild-type strain these properties were expressed only after growth in Fe-deficient medium. The mutation affects a gene responsible for regulating expression of the structural genes for the components of the high-affinity Fe uptake systems. The term fur, for Fe uptake regulation, is suggested for this new class of mutant.
