Weanling male mice were fed cyclohexanone and oxaldihydrazide, chemical precursors of cuprizone, without significant toxic effects. Supplementation of 0.5% cuprizone diets with thiamine, vitamin B12, and folic acid at level of 1000X dietary requirement and the diuretic (Diuril) was without significant antidotal effects. A multiple vitamin solution, riboflavin, and vitamin A reduced mortality over the experimental period but did not reduce the incidence of hydrocephalus or prevent the lesions of spongy degeneration. Copper at levels of 130 ppm and 260 ppm in feed was effective in reducing hydrocephalus incidence in mice fed 0.2% cuprizone. The copper supplements were without effect on the lesions of edema, spongy degeneration and astrogliosis. Mice became less likely to develop hydrocephalus as a result of cuprizone feeding as the age at which feeding began is increased; the incidence of hydrocephalus after 8 weeks of feeding was 100% when 5 week old mice were used, 75% when 6 week old mice were used, and 37.5% when 8 week old mice were fed. Hydrocephalus occurred in mice after 5 weeks of feeding 0.3% cuprizone; spongy degeneration occurred early, after 2 weeks, and glial proliferation was observed in most brains after 4 weeks.
