In this paper, a new biological layer-by-layer (LBL) cell sheet was developed for bone tissue engineering based on the interaction between hydroxyapatite micro-particles (mHAP) and MC3T3-E1 osteoblast cells. The LBL film mimicked a natural combination of inorganic and organic matrix. The properties of the cell sheets were characterized by live/dead assay, hematoxylin-eosin (HE) staining and scanning electron microscope (SEM) observation. The resulting cell sheet had exhibited well cell viability with the integrity of the extracellular matrix and mHAP particles. The interface between cell sheet and hydrogel showed that the living cells in deep layer have the same morphological structure as the cells on the superficial layer. The cell sheet could apart from the cell culture dish through deposited mHAP across in the middle of cells and the dish. In addition, the cell sheet maintained a high osteogenic capacity and could be taken as a more ideal bio-system for the repair of damaged bones without PIPAAm or any other ancillary equipment.
