Materials and MethodsOur strategy was to manipulate the GR population in vivo through ligand availability and hence autoregulation of the GR gene and in vitro by bolstering the endogenous population by introducing the GR gene into proximal tubule-like cultured kidney cells. In vivo experiments were performed on male Sprague-Dawley rats weighing 300-350 g. Adrenalectomies (ADX) were performed via the dorsal approach (Harlan, Indianapolis, Ind., USA) with the animals maintained on 0.9% NaCl prior to study. Experi-ments were always carried out within 12 days of ADX. Gc was administered in the form of a subdermal pellet releasing triamcinolone at a constant daily rate (Innovative Products, Toledo, Ohio, USA) of either 0.40 or 40 µg/100 g bw, low or high dose, respectively.
