Ultraviolet irradiation of the vegetal hemisphere of Rana pipiens eggs, beginning the first cleavage division, with doses greater than 8000 ergs/mm2resulted in the complete elimination of primordial germ cells in 100% of the larvae that were otherwise apparently normal. Irradiation of the animal hemisphere with equivalent doses of UV had no apparent effect on the formation of primordial germ cells. The effect of UV on germ cell formation was greatest at the time of the first division. After this, increasingly fewer eggs, which had been irradiated at the vegetal hemisphere, developed into larvae without germ cells. At the 8-cell stage, complete elimination of germ cells was not observed. Restitution of the germ line in irradiated eggs was obtained by injecting vegetal hemisphere cytoplasm from unirradiated eggs into the vegetal hemisphere of irradiated eggs, but restitution did not occur by injection of animal hemisphere cytoplasm. The UV action spectrum of germ cell inactivation suggests that the sensitive material is a nucleic acid. These results are consistent with the hypothesis of a specific cytoplasmic germinal determinant. This determinant is present only in the vegetal hemisphere cytoplasm; it is probably a nucleic acid; and it appears in embryonic development prior to the first cleavage division.
