The morphology of living amebae (Chaos chaos) during pinocytosis is described. The amebae were observed in a continuous flow compression chamber. Sections stained with the Periodic Acid Schiff Technique (PAS) and fluorescent antibody labeling techniques were correlated with the observations on the living amebae. Solutions of salts and proteins were tested for their activity in inducing pinocytosis. Experimental results showed that salts and proteins were active in inducing pinocytosis while non-charged solutes were not. Proteins were shown to be adsorbed to the plasmalemma prior to pinocytosis. The plasmalemma was shown by PAS technique to form the wall of the pinocytosis vacuole. A mechanism for pinocytosis is suggested which relates adsorption of charged solutes on the plasmalemma to a decrease in the tension and structural rigidity of the membrane. The weakened membrane is drawn down into the cytoplasm by its points of attachment to the contractile plasmagel system.
