DiscussionThe multiplication of mycoplasmas has been a matter of con troversy [5, 8]. Recent electron microscopic results have shown evidence for binary fission in Mycoplasma ho minis and a caprine mycoplasma strain [10]. Interpreting colony counts of filtered and UV-treated cultures, Furness et al.|(»| postulate a binary fission for M. pneumoniae. Rut so far there are no reports of continuous observations of living and multiplying mycoplasmas in a fluid environment. The results of such observations on M. pneumoniae as described here show binary fission (or division) to be the nor mal mode of multiplication. Furthermore they indicate a replica tion time of about 3 h, a time much shorter than the previously calculated It) h [12] or 6.5 h [6]. Media other than those used here might result in still shorter periods. The differences between these microscopic results and those obtained by colony counting could be due to the formation of ‘spherules’[9],‘clusters'[6] or ‘colonies’] 2] in the broth. These structures are preventing an exact evalua tion by colony counting of the true number of single organisms in broth.The mode of separation in the absence of any visible septa, and the disruption of the connecting string without loss of cellular material, cannot yet be explained. From preliminary electron microscopic studies (limit and Grimley, unpublished results) it seems possible that in'mature'filaments the cell content is con centrated within the knobs.
