MATERIALS AND METHODSMateriaZs. Highly purified hCG (CR-119; 12,800 IU/mg) was kindly provided by the National Institutes of Health, Bethesda, MD. Photographic emulsion L4 was frdm Ilford Ltd., Ilford, Essex, England. Isomethylbutyl-xanthine (IBMX) was obtained from Aldrich Chemical Co., Milwaukee, WI.Iodination of hCG. Iodination of hCG was performed by the lactoperoxidase method (32). The specific activity of the iodinated hormone was 30@ i/pg. Granulosa cell culture. Granulosa cells were collected from rat preovulatory follicles on the morning of proestrus by puncture and gentle expression into McCoy’s 5a (modified) medium containing 2 mM glutamine, 100 U/ml penicillin, 100 pg/ml streptomycin, and 20% fetal calf serum. Cells (lo6 cells/ml) were introduced into Falcon Optilux tissue culture dishes (35 x 10 mm) and cultured for 24 h as described by Nimrod and Lindner (37)
