The first genes coding for mouse and human tumor-rejection antigens recognized by T lymphocytes have now been isolated, and it is a likely prospect that several additional human genes will be identified in the near future. I shall attempt here to briefly summarize our findings and discuss how they lead to new possibilities for specific cancer immunotherapy. Our approach has been strictly limited to those antigens that are recognized by cytolytic T lymphocytes (CTL). This does not by any means reflect the belief that destruction by CTL is the only or even the most effective mechanism of tumor rejection. In fact, we have encountered several cases of very strong tumor rejection in the near-absence of detectable CTL (Boon and Kellermann, 1977; Boon and Van Pel, 1978). But CTL clones have proved extremely useful and reliable reagents that could support a genetic approach towards the analysis of tumor-rejection antigens (Weiss et al., 1980). Moreover, the demonstration that the immune receptor of CTL recognizes small peptides located in a groove of class-I major histocompatibility complex (MHC) molecules has made it possible to obtain a complete interpretation of thc genetic results
