Comparative studies are reported on the cytopathogenic effect of Brucella variants on guinea pig macrophages in tissue culture. Nine smooth (S) Brucella strains and 1 to 4 rough (R) variants derived from each S parent were studied. The findings indicate that S have little or no effect on infected host cells under the conditions employed. R strains, however, were found to be rapidly destructive to infected host cells as determined by the spectrophotometric measurement of the release of host cell constituents into the medium. No significant differences in the rate of growth of S and R in tissue culture was detected under the experimental conditions used and phagocytosis of S and R strains by the macrophages was found not to be essentially different. These observations, along with the finding that R strains are destructive to host macrophages within 6 hours, tends to support the view that destruction of host cells by R is not due to mechanical disruption.
