The production, isolation and properties of notatin from P. notatum are described in detail. Notatin is an enzyme of flavoprotein nature, catalyzing the oxidation of glucose to gluconic acid by means of atmospheric O2 with production of H2O2. A purified prepn. was buff to yellow in color, stable for at least 8 months at 0[degree], freely soluble in water, and will not pass through a nitro-cellulose membrane. Aqueous solns. have a pH of 6-7, and exhibit absorption maxima at 270/280, 375/380 and 450/460 mu. It inhibits Staphylococcus aureus in glucose broth in dilutions of 1:1 billion. This activity is rapidly destroyed at pH < 2 and > 8, or on heating to 60[degree]. The antibacterial activity is due to the production of H2O2 in the presence of glucose and O2. d-Xylose and d-mannose can function in the en-zymic reaction but glucose shows a higher activity. Maltose, arabinose and galactose show no O2 uptake.
