METHODSStudies were carried out in nine healthy mongrel dogs’(20-30 kg wt) with normal electrocardiograms. Prior to surgery, these anirnals were trained to lie quietly and unrestrained for several hours. The preparation used was generally similar to that previously reported from this laboratory(5, 11). Electromagnetic flow transducers were itnplanted on the circumflex branch and the ascending aorta. A pneumatic occlusive cuff (for obtaining temporary flow zero or for producing permanent coronary occlusion) was placed distal to the coronary flow transducer. In dogs I, 2, 4, and 6 a tube was implanted in the left circumflex branch distal to the cuff for measurement of peripheral coronary pressure and injection of radioactive xenon-133. Aortic pressure was measured by a catheter inserted into the aorta by way of the left common carotid artery or by direct implantation into the distal aortic arch. In dogs I, 3, 5, and 7 a tube was also placed in the coronary sinus for obtaining blood samples (11). The intracoronary tube was flushed once or twice daily with 20,000 U/ml of heparin while the other tubes were flushed daily with 1,000 USP U/ml of heparin. Fibrinolysin was occasionally needed to obtain free backflow of blood. In one dog, at the time of the implant procedures, the nerve connections to the heart were interrupted by a stripping technique. 2 Aortic and circunlflex phasic pressures were measured with P23Db and P23Gb Statham transducers, respectively. Records of phasic pressure and flows were recorded on an Electronics l In conducting the research described, the investigators adhered to the “Guide for Laboratory Animal Facilities and Care,” as promulgated by the Committee on the Guide for Laboratory Animal Facilities and Care of the Institute of Laboratory Animal Resources, National Academy of Sciences-National Research Council. 2 The authors are deeply indebted to Dr. David E. Donald, Mayo Clinic, Rochester, Minn., who was kind enough to separate the heart from its nerve connections at the time of implantations. Reference to this dog was made in a preliminary report on the effect of chronic cardiac denervation on the coronary circulation, at the American Heart Association Meeting, Miami, 1968 (Circulation, Suppl. 6: 88, 1968). Denervation appeared complete based on lack of response to intravenous tyramine and atropine during the experiment and at time of sacrifice the lack of response to cardiac nerve stimulation and the minimal myocardial catecholamine content.
